According to Vanity Fair, the New Establishment list 'ranks 50 innovators who have defined the year in terms of how we interact, how we work and play, and what cultural products we consume and how.'
At just 31-years-old, Zuckerberg is the youngest person to ever be ranked first on the list.
'At 31, Mark Zuckerberg stands out as something of an elder statesman,' says magazine editor Graydon Carter.
'He has not only proved himself as a brilliant entrepreneur; he may well become one of the great long-haul corporate leaders of the past half-century.
'The $240 billion company he scratched out of his dorm room at Harvard is apparently just his first step toward world domination in the realm of social networking.'
For his big cover shoot, Zuckerberg dressed in his everyday uniform of a gray tee shirt and pair of loose-fit jeans.
He did not, however, wear a hooded sweatshirt.
The cover line for Zuckerberg's article reads; 'He changed the world once. He says he'll do it again with Oculus Rift.'
Zuckerberg announced he had acquired Oculus VR - the maker of the virtual reality head-mounted display Rift - in a March 2014 Facebook post.
The Rift was created by 22-year-old Palmer Luckey and is expected to be released in early 2016.
'When you put it on, you enter a completely immersive computer-generated environment, like a game or a movie scene or a place far away,' said Zuckerberg of Rift back in 2014.
'The incredible thing about the technology is that you feel like you're actually present in another place with other people.
'People who try it say it's different from anything they've ever experienced in their lives.'
It has been a big year for Zuckerberg, with the billionaire tech titan announcing in July that he and his wife Priscilla were expecting their first child together - a baby girl.
There are also reports that Zuckerberg - who is estimated to be worth $36.8billion - recently purchased a $22million townhouse in New York City's trendy West Village neighborhood.
Others being named to this year's New Establishment list include Uber founder Travis Kalanick, Jeff Bezos of Amazon, comedian and writer Amy Schumer and TV host John Oliver.
The magazine also released a Powers That Be list that ranks visionaries who have 'consolidated their spheres of influence while continuing to affect the culture in new ways.'
That list is topped by their September cover star - singer Taylor Swift.
